摘要
目的:探讨正肝方及其拆方影响肝癌细胞增殖及端粒酶活性的特点及意义。方法:正肝方合方及其拆方中活血软坚类药物、益气养阴类药物、清热解毒类药物,分别制成水煎剂,灌喂正常大鼠,采血制备药物血清。以正常鼠血清为阴性对照,用药物血清温育B e l 7402人肝癌细胞。噻唑蓝比色法测定细胞增殖;聚合酶链反应酶联免疫吸附法(PCR EL ISA法)检测细胞端粒酶活性。结果:正肝方合方和方中活血软坚类药物、益气养阴类药物均可抑制B e l 7402细胞的增殖,清热解毒类药物不能抑制B e l 7402细胞的增殖。正肝方合方及方中活血软坚类药物、益气养阴类药物和清热解毒类药物均可抑制B e l 7402细胞端粒酶活性,且正肝方合方和活血软坚类药物的抑制作用优于清热解毒类药物。结论:正肝方可抑制B e l 7402细胞的增殖及其端粒酶活性,其抑制作用主要是通过方中活血软坚类药物和益气养阴类药物的作用来实现的。
Objective： To investigate the influence of Zhenggan recipe （正肝方） and its different combined herbs on the proliferation and telomerase activity of human hepatocarcinoma cells. Methods.. The herbs of Zhenggan recipe were divided into three groups ： the group of activating blood and softening hard mass （活血软坚）, the group of benefiting Qi and nourishing Yin （益气养阴）, and the group of clearing away heat and toxic materials （清热解毒）. These groups of herbs and Zhenggan recipe were prepared as decoction respectively, and fed to the normal rats. The drug sera were collected from the rats and incubated with cultured Bel - 7402 human hepatocarcinoma cells. While the normal rat sera were used as the control. Later, the proliferation of Bel - 7402 cells were observed by colorimetric assay （MTT）, and the telomerase activity of Bel-7402 cells was tested by polymerase chain reaction -enzyme linked immunoadsorbent assay （PCR -ELISA） method. Results： Zhenggan recipe, the group of activating blood and softening hard mass and the group of benefiting Qi and nourishing Yin could inhibit the proliferation of Bel - 7402 cells. The group of clearing away heat and toxic materials could not inhibit the proliferation of Bel - 7402 cells. All of the three groups and Zhenggan recipe could suppress the telomerase activity of Bel - 7402 cells. Zhenggan recipe and the group of activating blood and softening hard mass were better than the group of clearing away heat and toxic materials. Conclusion.. ghenggan recipe can inhibit the proliferation of Bel - 7402 cells. The inhibiting function of ghenggan recipe is realized by the functions of the activating blood and softening hard mass group and the functions of the benefiting Qi and nourishing Yin group.
