站台水环境应激对神经内分泌和胸腺细胞凋亡的影响及加味逍遥丸的调节作用 被引量:4

摘要 目的:探讨小站台水环境对小鼠血清糖皮质激素水平、胸腺5-羟色胺含量的变化及胸腺细胞的破坏、凋亡的影响,以及加味逍遥丸调节作用。方法:实验于2003-03/12在江西中医学院附属医院临床检验中心完成。选择10月龄昆明种小鼠,随机分为14组:应激72h组;应激72h给药组;应激48h组;应激48h给药组;应激24h组;应激24h给药组;应激12h组;应激12h给药组;应激8h组;应激8h给药组;应激4h组;应激4h给药组;正常对照组;正常对照给药组。每组10只。利用小站台水环境诱发慢性持续性应激模型,观察不同应激时间小鼠站台前后胸腺质量及组织指数、血清糖皮质激素含量、胸腺5-羟色胺含量、胸腺细胞凋亡状况,以及加味逍遥丸在不同时间不同层次的调节作用。结果:参加实验小鼠140只,全部进入结果分析。①胸腺质量和脾质量:应激24h,48h,72h组均比对照组显著降低(P<0.001或P<0.01);应激72h给药组与相对应应激组相比显著升高[(71.34±10.52),(57.75±9.17)g,P<0.01];[(70.28±9.05),(58.75±7.72)g,P<0.01];应激48h给药组脾质量与相对应应激组相比也显著升高[(75.51±5.05),(64.75±6.22),P<0.01]。②胸腺指数和脾指数:应激48h、72h组小鼠显著低于对照组(P<0.001或P<0.01)。应激72h给药组与相对应应激组相比增高[(3.02±0.42),(2.21±0.42),P<0.01];[(2.97±0.12),(2.36±0.34),P<0.01]。③小鼠血清糖皮质激素含量:应激12h,24h,48h,72h组与对照组相比明显提高(P<0.001或P<0.01),应激72h给药组与应激72h组相比显著降低[(19.16±5.06),(23.29±4.64)g/L,P<0.01]。④小鼠胸腺组织、脾组织5-羟色胺含量:应激8h,12h,24h,48h,72h组胸腺5-羟色胺含量比对照组显著提高((P<0.001或P<0.05);应激48h,72h组脾组织5-羟色胺含量比对照组显著提高(P<0.001或P<0.01),应激72h给药组与相对应激组相比有所降低[(2.15±0.31),(2.91±0.25)μg/g,P<0.001];[(6.05±2.08),(7.96±1.70)μg/g,P<0.05]。⑤荧光显微镜观察TFAR19� AIM: To investigate the effect on the level of glucocorticoid in serum, change of the content of 5-hydroxytryptamine (5-HT) in thymus and destroy and apoptosis of thymus by platform-over-water stress, and study the regu- lative role of Jiawei Xiaoyao pill. METHODS: The experiment was completed in the Clinical Laboratory Center, Affiliated Hospital of Jiangxi College of Traditional Chinese Medicine between March and December 2003. Kunming mice of 10 months old were randomly divided into 14 groups with 10 mice in each group: 72- hour stress group; 72-hour.stress and treatment group; 48-hour stress group; 48-hour stress and treatment group; 24-hour stress group; 24-hour stress and treatment group; 12-hour stress group; 12-hour stress and treatment group; 8-hour stress group; 8-hour stress and treatment group; 4-hour stress group; 4-hour stressand treatment group; normal control group; normal control treatment group. Models of chronic continuous stress were in- duced by the platform-over-water environment, the changes thymus mass, thymus index, level of glucocorticoid in serum, 5-HT content in thymus and thymus apoptosis at different stress time were observed, and the regulative effect-of Jiawei Xiaoyao pill at different time and different stratification was detected. RESULTS: All the 140 mice were analyzed in the results. ①Thymus and spleen masses: The masses in the 24, 48 and 72-hour stress groups were signif- icantly decreased as compared with those in the control group (P 〈 0.001 or 0.01); The masses in the 72-hour stress and treatment group were significantly increased as compared with those in the 72-hour stress group [(71.34±10.52), (57.75±9.17) g, P 〈 0.01; (70.28±9.05), (58.75±7.72) g, P 〈 0.01]. The spleen mass in the 48-hour stress and treatment group was significantly increased as compared with that in the 48-hour stress group [(75.51±5.05), (64.75±6.22) g, P 〈 0.01]. ②Thymus and spleen indexes: The indexes in the 48 and 72-hour stress groups w
