辛伐他汀对单侧输尿管梗阻大鼠肾间质纤维化的影响

摘要 目的研究羟甲基戊二酰辅酶A(HMG-COA)还原酶抑制剂(HRI)辛伐他汀(simvastatin)对输尿管梗阻大鼠(UUO)肾间质纤维化的影响及可能机制。方法通过结扎单侧输尿管造成大鼠肾间质纤维化,随机分为假手术组、模型组和给药组,术后9天处死各组大鼠,取梗阻侧肾做组织学检查,HE染色,并用免疫组化方法检测肾组织中结缔组织生长因子(CTGF)、α-平滑肌肌动蛋白(α-SMA)、胶原-Ⅳ(COL-Ⅳ)的水平。结果模型组出现了明显的间质纤维化,给药组肾间质纤维化与模型组相比有所减轻,CTGF、α-SMA、COL-Ⅳ的水平也明显减低。结论辛伐他汀可以影响细胞因子、抑制细胞增殖、减轻细胞外基质的积聚,从而减轻间质纤维化,发挥肾保护作用。 Objective To evaluate the effect and possible mechanism of HMG-COA reductase inhibiter simvastatin on interstitial fibrosis of unilateral ureteral obstruction (UUO) rats. Methods The tubulointerstitial fibrosis model was established by unilatered ureteral obstruction. These rats were randomly divided into sham-operated , UUO and simvastatin treatment group. These rats were sacrificed at 9 days after UUO or sham-surgery. The morphological changes were observed.The expression of CTGF, α-SMA, COL-Ⅳ in the obstructed kidney was assessed by immunchistachemistry methods. Results The levels of CTGF, α-SMA, COL-IV in UUO group increased remarkably comparing with the sham-operated group. In comparison with the model group, the levels of CTGF, α-SMA, COL-Ⅳ were significantly reduced by simvastatin treatment. Conclusions Simvastatin may suppress interstitial fibrosis by effect cell factor, suppress cell proliferation, reduce extracellular matrix accumulation.
