摘要 目的:建立γ-干扰素(IFN-γ)基因表达的绝对定量实时PCR检测技术。方法:应用实时定量PCR方法检测25名健康运动员生理状态下外周血白细胞IFN-γmRNA的表达。主要结果和结论:应用实时定量PCR方法能够对健康受试者外周血白细胞自发性IFN-γmRNA表达进行定量分析,相关系数r>0.99。 Objective To develop a method for quantitative real- time detection of interferon- γ (IFN - γ) using polymerase chain reaction (PCR). Methods Gene expression of IFN - γ in peripheral blood mononuclear cell (PBMC) from 25 healthy individuals was detected by PCR. An endogenous “housekeeping” gene GAPDH was used to normalize the results. Results and Conclusion The spontaneous IFN- γ mRNA expression in PBMC in a given sample could be measured by PCR with relativity correlation coefficient of r 〉 0.99.
