土贝母制剂诱导Jurkat细胞凋亡及其对细胞周期的影响

摘要 目的探讨土贝母制剂对Jurkat(人T细胞白血病细胞株)细胞增殖抑制率及对细胞凋亡,细胞周期各时相的变化。方法应用MTT比色法和流式细胞仪检测经士贝母制剂对Jurkat细胞增殖抑制率的时间效应和浓度效应及凋亡率的变化。结果土贝母制剂对Jurkat细胞有明显的抑制作用,且呈剂量依赖性,流式细胞仪分析,G1期细胞增高,S期细胞减少,G2/M期细胞增多,凋亡率上升。结论土贝母制剂能够抑制Jurkat细胞增殖,抑制G1期细胞向S期转化的进程,促进Jurkat细胞凋亡。 Objective To investigate the influence of Bolbcstemma Paniculatum preparation on inhibitory rate of cell proliferation, cell apoptodsis and changes of every cell cycle stage. Methods MTT colorimetric assay and flow cytometry(FCM) wereusedto measure the change of time effect, concentration effect and apoptotic rate by Bolbcstemma Paniculatum preparation on inhibitory rate of Jurkat cell proliferation. Results Bolbcstemma Paniculatum preparation can obviously inhibit the Jurkat cell, and this effect is dependent on the dosage. Analyzed by FCM, we find that C1 phase cells increased, S phase cells decreased, C2/M phase cells increased and apoptotic rate raised. Conclusion Bolbcstemma Paniculatum preparation can inhibit the proliferation of Jurkat cell, inhibit the transformation from C1 phase cells to S phase cells, and accelerate the apoptosis of Jurkat cell.
