摘要 目的:探讨血管生成抑制素和反应停对淋巴管内皮细胞增殖和游走的影响及作用机制。方法:淋巴管内皮细胞取自猪的胸导管。采用划线法和MTT法观察血管生成抑制素和反应停对淋巴管内皮细胞增殖和游走的影响。Hoechst染色检测细胞凋亡。结果:划线法和MTT法均显示血管生成抑制素和反应停对淋巴管内皮细胞的增殖和游走有明显的抑制作用。Hoechst染色证实,经血管生成抑制素和反应停处理后的淋巴管内皮细胞,核周围有凋亡小体。结论:血管生成抑制素和反应停对淋巴管内皮细胞的增殖和游走具有明显的抑制作用,且有剂量依赖性。血管生成抑制素和反应停具有促进细胞凋亡的作用。 Objective: To study the effect and mechanism of angiostatin and thalidomide on proliferation and migration of lymphatic endothelial cells (LEC). Methods: LEC from pig thoracic duct were cultured. Scraping line method and MTT were used to observe the effect of angiostatin and thalidomide on proliferation and migration of LEC. Hoechst staining was applied to examine apoptosis of LEC. Results: Angiostatin and thalidomide evidently inhibited proliferation and migration of LEC (P〈0. 01). Hoechst staining showed apoptotic body surrounding the nucleus of LEC after treated by angiostatin and thalidomide. Conclusion: Angiostatin and thalidomide can evidently inhibit the proliferation and migration of LEC in a dose-dependent manner and they can also induce apoptosis of LEC.
