摘要
目的:建立一种方便、实用的慢性器官移植排斥反应模型,为研究器官移植后慢性排斥反应的发病机制、预防和治疗提供平台。方法:以近交系F344大鼠和近交系Lewis大鼠作为供、受体,取供体的双肾作为供肾,借助静脉支架管,行供体的静脉与受体肾静脉的端端吻合,肾动脉与腹主动脉行端侧吻合。施行原位异体肾移植。移植后即给与环孢素腹腔注射。并于术后4周后行病理检查。结果:所有手术均于60min内完成。所有的受体均存活超过60天,围手术期死亡数为零。受体无急性排斥反应发生。4周后可见典型的慢性排斥反应发生。结论:建立了大鼠慢性排斥反应肾移植模型。为研究慢性排斥反应的发生、进展和干预治疗提供了良好的平台。
Objective: To establish an economic and feasible rat renal transplantation model of chronic rejection. Methods: Inbred F344 and Lewis rats were used as the donors and the recipients. Both kidneys were removed from the donors. With the help of PTCA tube, an end-to-end anastomosis of vein was operated .After transplantation,recipients were received ciclosporin every day. Results: All operations were finished within 60 min. No recipients died during operation. No acute rejection evidence was seen. Chronic rejection could be seen obviously 4 weeks after operation. Conclusion: The newly established method is feasible and reliable. The good model of studying chronic rejection were offered.
