摘要
采用造血祖细胞体外培养和造血生长因子检测等实验血液学技术,研究三七总皂甙对小鼠红系祖细胞增殖调控的生物学机理.结果表明:三七总皂甙对正常或骨髓抑制一贫血模型小鼠的红系祖细胞增殖有明显促进作用;三七总皂甙亦可提高阿糖胞苷所致祖细胞的“自杀”率;经三七总皂甙诱导制备的脾细胞、L细胞(成纤维细胞株)培养上清液对红系祖细胞的增殖具有较高刺激活性.该文提示三七总皂甙可能通过诱导造血微环境中的成纤维细胞、淋巴细胞等分泌较高活性的红系造血调控因子和/或协同造血生长因子,以促进红系祖细胞增殖分化,这或许是三节“补气、活血”的生物学机理之一.
By using the culture of hematopoietic progenitor cells, and hematopoietic growth factor assay, the effect of total saponins of panax notoginseng (TSPN) on the proliferation of erythroid progenitors and its mechanism were studied. The results indicated that TSPN augmented the proliferation of BFU-E and CFU-E, that the spleen cell conditioned media, prepared by treating spleen cells with TSPN, had relatively strong stimulation on the proliferation of BFU-E, and that the cultured L cell supernates, prepared by adding TSPN in cultured media in vitro, increased the formation of BFU-E and CFU-E. Our research suggests that TSPN may enhance erythropoiesis by activating directly and/or indirectly fibroblasts and lymphocytes in hematopoietic inductive microenvironment to produce some hematopoietic growth factors. This may be one of the mechanisms for the antianemic effect of panax notoginseng.
