摘要
柱状田头菇、鲍鱼菇、红菇子实体浸出液经硫酸铵分级沉淀、活性炭柱吸附、DEAE-SepharoseFF离子交换层析及Sephadex G-100分子筛层析,得到纯化的柱状田头菇凝集素(Agrocybe cylindraceaLectin,简称ACL)、鲍鱼菇凝集素(Pleurotus abalonusLectin,简称PAL)、红菇凝集素(Russula depallensLcetin,简称RDL).从纯化结果来看,RDL的纯化倍数及蛋白回收率比ACL和PAL高.ACL和RDL对供试动物的红细胞都有凝集作用,PAL只凝集部分动物红细胞并表现出血型专一性.ACL、RDL和PAL除凝集红细胞外也能凝集动物的其它细胞,但PAL不凝集S180瘤细胞.此外,3种凝集素也能凝集某些微生物,如细菌、放线菌、植物病原菌.凝集结果表明ACL、RDL的凝集活性普遍高于PAL.
The extractions of Agrocybe cylindracea Lectin (ACL) , Pleurotus abalonus Lectin (PAL) and Russla depallens Lectin (RDL) were purified by ammonium sulfate precipitation, active carbon adsorption, DEAE-Sepharose FF ion exchange chromatography and Sephadex G-100 gel chromatography sequently, and Agrocybe cylindracea Lectin (ACL), Pleurotus abalonus Lectin (PAL) and Russla depallens Lectin (RDL) were collected respectively. The results showed that the purified folds and the protein recycle rate of RLL higher than others. Both ACL and RDL have agglutination on erythrocyte of tested animals. However PAL can only agglutinate erythrocyte in parts of animals and has speciality to blood type. In addition to agglutinate the erythrocyte, ACL, RDL and PAL can also agglutinate the other cells of the animal, but PAL cannot agglutinate S:80 tumor cell. Furthermore all three kinds of lectins can also agglutinate some microorganisms, such as bacterium, actinomyces and plant pathogenic bacterium. The results of agglutination shows that the agglutinative activities of ACL and RDL normally higher than the PAL's
