摘要
大鼠颌下腺为表皮生长因子(EGF)的主要来源之一。EGF可刺激垂体分泌ACTH。将大鼠分组为针刺与唾腺相关之“腮腺”耳穴组,针刺与垂体相关之“脑点”耳穴组和不施加针刺的对照组。测定痛阈并分别以原位杂交和免疫组织化学技术探测颗粒曲管(GCT)细胞的EGFmRNA、EGF受体(EGFR)-免疫反应性(IR)及EGF-IR和垂体ACTH-IR。结果显示针刺后EGFmRNA及EGFR-IR提高;而EGF-IR在两个耳针组变动均不显著;垂体ACTHIR仅在“脑点”组减弱,并且在ACTH变动与镇痛效应之间呈负相关。
The rat submaxillary gland is one of the main sources of EGF which can stimulate the pituitary to secrete ACTH. The rats were grouped into the needling at the earacupoint 'Sai-xian' correlated with the salivary glands and at the earacupoint 'Nao-dian' correlated with the pituitary, and the control treated with no neddling. The pain threshold was measured, and EGF mRNA,EGFR (receptor)-IR (immunoreactivity) and EGF-IR in granular convoluted duct (GCT) cells and the pituitary ACTH-IR were detected by in situ hybridization and immunohistochemistry techniques respectively. The results showed that after acupuncture the EGF mRNA and EGFR-IR were enhanced,while the alteration of the EGF-IR was insignificant in both earacupuncture groups; the pituitary ACTH-IR was attenuated (by promoted secretion) with a correlation between the alteration of ACTH-IR and analgesic effect only in the 'Nao-dian' group. The relative specificity in correlation between the 'exterior' (acupoint) and 'interior' (hormone, growth factor) was discussed.
