摘要
采用6-氨基喹啉-N-羟基琥珀酰亚胺基氨基甲酸酯(AQC)为衍生剂,与各种氨基酸柱前定量衍生后,用WatersHPLC仪,AQC-Tag ̄TMC_18柱,以140mmol/L从醋酸钠溶液(pH=4.95)为溶剂A,乙腈-水(3:2)为溶剂B进行梯度洗脱,检测波长为248nm,以α-氨基丁酸为内标。所有18种氨基酸在37min内测定完毕,各种氨基酸测定的重现性RSD%均小于2.0%(n=5),平均回收率为97.0%6~105.0%(n=5)。
The paper reports the determination of amino acids in compound amino acid injections using HPLC-AccQ-Tag method. All the amino acids reacted with 6-aminoquinoly-N-hydroxysuccinimdyl carbamate(AQC)respectively and quantitatively and produced corresponding derivatives which were analized by HPLC.Chromatographic conditions were AccQ-TagTM C18 for amino acids analysis(3.9mm×15cm),mobile phase of program elution (A)sodium acetate solution(pH=4.95)(B) acetonitrilewater(3:2);using α-aminobutyric acid as internal standard and detected at 248nm by UV-detector.The method was accurate,rapid,simple, convenint and high sensitive(×10-9mol). Eighteen kinds of amino acids were completely determined in 37 minutes. The correlation coefficients of the calibration graphs for seventeen amino acids were good (r≥0.9993), RSD of all amino acids were less than 2.0%and the average recoveries range were 97.0%~105.0%(n=5).
