摘要
目的:考察吲哚美辛生物黏附微囊的各种性质.方法:考察吲哚美辛生物黏附微囊在大鼠体内外的黏附性能;通过扫描电镜、差示热分析手段研究吲哚美辛生物黏附微囊的外观形态及药物在微囊中的存在形式.结果:吲哚美辛生物黏附微囊粒径在750~850 μm范围内占89%,微囊在大鼠小肠体外及体内的平均滞留率分别为90%和80.8%,大鼠口服微囊2.5 h后处死,在胃和小肠中的平均滞留率分别是11.2%和85.3%.差示热分析结果表明,吲哚美辛在微囊中仍以结晶形式存在.结论:吲哚美辛生物黏附微囊外观圆整,粒度分布均匀,大鼠体内外黏附性能优良.
OBJECTIVE To investigate the properties of indomethacin biaoadhesive microcapsules. METHODS Indomethacin microcapsules were examined in vivo and in vitro in terms of their bioadhesive characteristics to the mucosa of rat small intestine. The appearances of microcapsules were observed with scanning electron microscope and the form of Indomethacin in microcapsules was studied by DSC analysis. RESULTS The percent of microcapsules with a diameter of 750-850μm was 89%. The average retention rates of microcapsules in vivo and in vitro in the rat small intestine were 90% and 80. 8% respectively. The average retention rates of microcapsules in the stomach and in the small intestine were 11.2% and 85.3% respectively after 2. 5 h administrated by oral in rat. The results of DSC analysis showed that Indomethacin still existed in crystal form in the microcapsules. CONCLUSION The bioadhesive microcapsules are spheric and uniform, and have a good adhesive ability both in vitro and in vivo in rats.
