摘要
目的探讨紫杉醇对喉癌细胞Hep-2生长抑制作用;紫杉醇对细胞角蛋白基因13(cytokeratin13,CK13)mRNA表达的影响。方法应用MTT法检测紫杉醇对Hep-2细胞的抑制率;采用半定量RT-PCR法检测紫杉醇作用Hep-2细胞后CK13mRNA的表达。结果紫杉醇在10-7mol/L以上浓度时,对Hep-2细胞抑制率大于30%(P<0.01);10-8mol/L以上浓度的紫杉醇可下调CK13mRNA的表达,其作用呈剂量依赖性。结论Hep-2细胞对10-7mol/L以上浓度紫杉醇具有高度敏感性;紫杉醇可能通过抑制CK13mRNA表达从而抑制肿瘤的侵袭及转移。
[Objective] To investigate the effects of paclitaxel on CK13 gene expression in larynx carcinoma cells, and the possible underlying mechanism. [Method] The effects of paclitxel on the proliferation of Hep-2 cells in vitro was determined by MTT(Thiazolyl blue)assay. The expression of CK13 mRNA in Hep-2 cell were assayed by RT-PCR. [Results] The IC50 of paclitaxel in Hep-2 cell were 6.25×10^-7 mol/L.The difference of growth inhibitory rate between 104 mol/L paclitaxel and 10^-7 mol/L paclitaxel was significant (P 〈0.01). Paclitaxel could down-regulate the expression of CK13mRNA when the concentration was 〉/10^-8mol/L. [Concblsion] When the concentration is ≥ 10^-7 mol/L,Paclitaxel can inhibit the expression of CK13mRNA.Larynx carcinoma cell Hep-2 is sensitive to paclitaxel when the concentration is ≥10^-7 mol/L.Paclitaxel may restrain the invasion and transfer of carcinoma in the way of inhibit the expression of CK13mRNA.
