PTEN和Bcl-2基因在骨肉瘤组织中的表达及意义

摘要 目的:探讨PTEN,B cl-2基因在骨肉瘤组织中表达的意义和和临床价值。方法:用SP免疫组化方法检测PTEN和B cl-2在62例骨肉瘤及20例骨软骨瘤表达情况结果PTEN的阳性表达率在骨肉瘤中为58.1%(36/62),在骨软骨瘤中为85%(17/20),两者差异显著(P<0.05);B cl-2的阳性表达率在骨肉瘤中为51.6%(32/62),在骨软骨瘤中为20%(4/20),两者差异显著(P<0.05)。PTEN的低表达与分化程度无关与肺转移有关,B cl-2的高表达与分化程度有关。术后生存5年以上者的PTEN蛋白阳性率(66.7%)较生存时间不超过5年者(28.1%)高(P<0.05)。骨肉瘤组织中B cl-2与PTEN基因的表达未见显著性相关(P>0.05)。结论:骨肉瘤中存在PTEN呈低表达而B cl-2高表达,PTEN低表达和B cl-2高表达于骨肉瘤侵袭、转移有关。 Objective:To investigate the significance and the clinical value of the expression of PTEN , Bcl- 2 in osteosarcoma. Methods:The expression of PTEN ,Bcl-2 had be detected for the specimens of 60 osteosarcoma and 20 osteochondroma with immunohistochemistry S-Pmethod. Results:the positive rate of PTEN was 58.1% (36 /62) in osteosarcoma group and 85% (17/20) in osteochondroma group, there was a prominent difference between them(P〈0.05), the positive rate of Bcl-2 was 51.6%(32/62) in osteosarcoma group and 20%(4/20) in osteochondroma group. there was a prominent difference between them(P〈0. 05). PTEN was related with pulmonary metastasis instead of differentiation(P〈0.05) ;Bcl-2 was related with differentiation(P〈0.05). PTEN expression showed significantly stronger in the patients whose survival time over 5 years(66.7%)than in the patients whose survival time was less than 5 years (28. 1 % )(P〈0. 05 ). Conclusion :There existed low expression of PTEN and over-expression Bcl- 2 proteins . Coexpression of these antioncogenes played an important role in invasion and rnatastasis in osteosarcoma.
