摘要 目的:建立用高效液相色谱法(HPLC法)测定人血浆样品中阿德福韦浓度的定量方法。方法:应用Agilent 1100高效液相色谱仪,以Eclipse XDB C18柱为色谱柱(150 mm×4.6 mm,5μm),流动相为0.2%H3PO4-甲醇(87:13),检测波长为260 nm,流速为1.0 mL/min。结果:阿德福韦浓度在20-2 000 ng/mL范围内与峰面积线性关系良好(r=0.999 3)。结论:HPLC法简便、快捷、灵敏、准确,适用于阿德福韦药代动力学研究. Objective: To establish HPLC method for assaying adefovir in human plasma. Methods: Adefovir in the samples was quantitatively analyzed by HPLC with the external standard. HPLC conditions for adefovir were as follows: Agilent 1100 HPLC instrument was used with C18 column(150 mm × 4. 6 mm,5 μm). The mobile phase consisted of 0. 2% H3PO4 -CH3OH in plasma with the flow rate 1.0 mL/min. They were determined at UV wavelength of 260 nm. Re, suits: The nicer linear range was 20-2 000 ng/mL of adefovir concentration(r=0.999 3). Conclusion: HPLC is simple, rapid, accurate, and suitable for application to study on pharmacokinetics of adefovir.
