垂体腺瘤的受体靶向性基因治疗研究

摘要 目的构建并评价受体靶向性基因导入系统对垂体腺瘤的靶向性治疗作用。方法构建GE7系统介导的基因治疗系统,通过垂体腺瘤GH3细胞及对照细胞的体外基因转染实验,观察其对垂体腺瘤的转染能力和靶向性。结果成功构建GE7基因治疗系统;基因转染后,GH3细胞特异性表达报告基因,并被治疗基因特异性杀伤。结论GE7基因治疗系统能够达到靶向性治疗垂体腺瘤的目的。 Objective To construct and evaluate the receptor targeted gene therapy system for pituitary adenoma. Methods The GE7 - system-mediated gene therapy system was constructed. Pituitary tumor cell line GH3 cells and control cells were treated with the gene therapy system. The gene transfection efficiency and targeting of the therapy system were observed. Results The GE7 system-mediated gene therapy system was constructed successfully. After gene transfection, GH3 cell specifically expressed reporter gene and killed specifically by therapy gene. Conclusion GE7 system-mediated gene therapy system can transfer target gene to pituitary adenoma effectively and specifically.
