摘要 目的探讨COX-2选择性抑制剂NS398(氮-2,环己氧-4,硝基苯甲基磺胺)是否对小鼠皮肤光老化有延缓作用。方法采用8甲氧补骨脂素及长波紫外线(8MOP/UVA)共同作用于小白鼠,进行皮肤光老化动物模型的造模,将NS398注射到实验动物真皮层干预皮肤光老化过程,免疫组织化学染色法、HE染色和弹力纤维染色(Weigert法)分别对与皮肤衰老有关的TIMP1,P53和PCNA表达、皮肤组织结构和弹力纤维的改变进行分析。结果注射NS398的实验组P53的含量减少,TIMP1和PCNA的含量增加,与皮肤光老化有关的皮肤组织结构和弹力纤维的改变明显减轻。结论NS398对小鼠皮肤光老化有延缓作用,可能为临床抗皮肤衰老提供了一条新途径。 Objective To investigate the preventing effect on mice's skin photoaging of COX-2 selective inhibitor,NS398. Methods In this experiment ,we used 8-MOP and UVA to act on the skin of mice back to set up an animal model of skin photoaging. Injected NS398 to the dennis of mice back to intervene the process of skin photoaging. The expression of TIMP-1 ,P53 and PCNA ,the changes of skin structure and elastic fiber were determined with immunohisochemistry, HE strains and elastic fibre strains. Results Treatment with NS398 prevented the increase of P53 and the decrease of TIMP-1 PCNA protein. NS398 could also inhibit the senescence-associated changes of skin structure and elastic fiber. Conclusion NS398 can prevent mice's skin photoaging. It may provide a new way to prevent skin photoaging.
