尿素交连明胶对正常人体免疫系统细胞因子的影响

摘要 目的探讨不同浓度的3.5%尿素交连明胶(UG)对正常人体免疫系统细胞因子的影响。方法选择健康志愿者12名,严格无菌条件下抽取静脉血15 ml,肝素抗凝。计数后将细胞平均分成五组:1640培养液阴性对照组(C1)、10%血浆对照组(C2)、10%UG观察组(H1)、20%UG观察组(H2)、40%UG观察组(H3)。各组分别在未加药(T1)、加脂多糖(LPS,T2)、加植物血凝素(PHA,T3)三种条件下,使单个核细胞在不同的培养液中培养48 h后,以酶联免疫吸附法(ELISA法)检测白细胞介素-2、10(IL-2、IL-10)和干扰素-γ(IFN-γ)的浓度。结果各组IL-2I、L-10、IFN-γ在T1、T2、T3组间比较无显著性差异(P>0.05);IL-2和IFN-γ在T1与T2两种条件下比较无显著性差异(P>0.05),但两者与T3条件下比较有显著性差异(P<0.05);IL-10在三种条件下比较均有显著性差异(P<0.05)。结论UG在体外试验中不抑制正常人体免疫系统机能。 Objective To study the influence of Haemaccel ( colloid plasma substitute ) with different concentrations on IL-2, IL-10 and IFN-γ in healthy persons. Methods 15 ml venous blood from 12 healthy volunteers was taken and single nuclear cells were separated from the blood. After counting the cells, the cells were divided into five groups: 1640 culture medium negative controlling group(C1 ), 10% plasma group(C2 ), 10% Haemaceel group( H1 ), 20% Haemaeeel group( H2 ) and 40% Haemaeeel group(H3 ). Then each group was subdivided into no adding terms(T1 ), adding LPS terms(T2 ) and adding PHA terms(T3 ). The cells were cultured in each culture medium for 48 hours. Enzyme-linked immunosorbent assay (ELISA) was used to detect the level of IL-2, IL-10 and IFN-γ. Results The levels of IL-2 and IFN-γ were significantly higher in adding PHA group (P〈0.05). There was a significant defference in IL-10 among negative controlling group, adding LPS group and adding PHA group (P〈0.05). Conclusion Haemaccel in vitro has no influence on the immune system of healthy persons.
