摘要
目的观察托吡酯(TPM)对杏仁基底外侧核电点燃癫痫大鼠海马半胱天冬氨酸蛋白酶(Caspase-3)mRNA表达的影响.方法将双极电极插入大鼠左侧杏仁基底外侧核,给予慢性电刺激使大鼠达到点燃状态,观察其发作过程并记录脑电图.采用半定量RT-PCR法检测托吡酯治疗组和对照组电点燃大鼠海马组织的Caspase-3 mRNA表达.结果大鼠平均(13.50±3.99)次刺激达到点燃,记录到后发放时程在21 450~119 720 ms之间.电点燃大鼠海马Caspase-3 mRNA表达明显升高,托吡酯治疗组的Caspase-3 mRNA表达下调.结论在大鼠电点燃癫痫模型中,Caspase-3可能参与了点燃过程,托吡酯可抑制癫痫过程中神经元的凋亡.
Objective To observe the effect of topiramate on the expression of Caspase-3 mRNA in the basolateral amygdala electrical (BLA) kindled epilepsy rats hippocampi. Methods Bipolar electrodes were implanted in BLA in rats, through which the rats received chronic electrical stimuli for kindling. Their seizure processes and behavior were observed, and electroencephalogram (EEG) was recorded during kindling and after kindled. At the end, the hippoeampi were extracted and Caspase-3 mRNA in the hippocampi were analyzed with semi-quantitative RT-PCR Results Mean stimulus needed for kindling was (13.50±3.99) times. Their afterdischarge duration (ADD) recorded was between 21,450 ms and 119,720 ms. Significant upregulation of Caspase-3 mRNA was found in kindled rats' hippocampi, while expression of Caspase-3 mRNA was decreased in topiramate treating groups. Conclusion In seizures induced by electrical kindling, Caspase-3 possibly participates in the process of kindling Topiramate might inhibit the apoptosis of the neurons in epilepsy.
