高压芒刺静电场对小鼠免疫共刺激分子表达的影响

摘要 目的:通过时程/量效观察不同强度高压芒刺静电场对免疫细胞表面共刺激分子表达的影响.方法:采用不同强度高压芒刺静电场对小鼠进行全身辐照,流式细胞术检测其胸腺细胞CD2和腹腔巨噬细胞CD48表达的变化.结果:低电压(10~20 kV)处理组的CD2及CD48表达量显著高于0 kV组(P<0.01);而高电压(35 kV)处理组的表达量明显低于0 kV(P<0.01);低电压时程的结果表明,处理后2 h CD2和CD48表达量开始增加,4~8 h达到高峰,与0 kV组比较差异具有非常显著性(P<0.001);高电压的时程结果表明,于处理后2~4 h CD2和CD48表达量即达到最低值,之后逐步回升(P<0.001),但到24 h未能恢复到对照组水平.结论:不同强度高压芒刺静电场可引起免疫共刺激分子表达量不同的生物效应,在免疫调节反应中发挥重要功能. Objective To observe the time-course and dose-effects of different doses of high voltage prick electrostatic field (HVPEF) on the expressions of CD48 in peritoneal macrophages and CD2 in thymocytes. Methods Whole bodies of mice were treated with HVPEF. Immuno-fluorescent FCM was adopted to detect the changes of CD48 and CD2 expressions. Results CD2 and CD48 expressions increased significantly after treated with lower voltages (10-20 kV) (P〈0. 01), while higher voltage (35 kV) decreased expressions of CD2 and CD48 (P〈0.01) ; CD2 and CD48 expressions began to increase 2 h and to reach its peak at 4-8 h after treated with 15 kV (P〈0. 001) ; CD2 and CD48 expressions decreased to their minimums 2-4 h after treated with 35 kV, then recovered gradually (P〈0. 001). Conclusion The different doses of HVPEF might cause different biological effects, which suggests HVPEF has potential immune-regulatory effects and might play an important role in immune modulation.
