摘要
通过酶解获得了糖海带、掌状海带和裙带菜孢子体原生质体。比较了酶的种类和浓度、酶液氢离子浓度及渗透压、高渗预处理等对原生质体产量的影响。原生质产量最高可达3×108个/g鲜重。达到了无菌培养和细胞壁再生。
Protoplasts were respectively isolated from sporophytes of Laminaria saccharina L. digitata and Undariapinnatifida through enzymatic removal of cell wall. On optimizing conditions such as the kind and concertration ofenzymes ,PH value and osmotic pressure of enzyme solutions,etc,the highest vield was 3 × 108 protoplasts /g FW.Axenic cultures were obtained and cell wall regenerated during subsequent culture.
