摘要
设置养分贮存损失状态不同的4类培养液,分别添加一定浓度梯度的L-谷氨酰胺(L-G lu tam ine,L-G ln)培养大鼠前体脂肪细胞。通过形态学观察、M TT比色、油红O染色提取法,比较各组细胞形态及增殖与分化的效果。结果表明,添加0 mm o l/L L-G ln的新鲜培养液和配制后4℃冷藏20 d,再添加0.685 mm o l/L L-G ln的培养液,均较利于前体脂肪细胞的增殖与分化;配制后4℃冷藏90 d的培养液,对前体脂肪细胞培养效果较差,添加L-G ln也不能使培养效果得到改善。另外,启封后的M 199和胎牛血清(F eta l C a lf Serum,FCS)分别冷藏不宜超过90 d。说明及时满足培养液中L-G ln的需求量,可使前体脂肪细胞达到较好的增殖与分化状态。
L-Gln was added to the 4 culture media of different nutritions for rat preadipocytes. The proliferation and differentiation of preadipocyte were determined by MTT spectrophotometry and Oil Red O staining respectively. The results show that the addition of 0 mmol/L L-Gln in fresh culture media and 0.685 mmol/L L-Gln in the culture media which had been stored at 4℃ for 20 d benefited proliforation and differentiation of the preadipocytes. When culture media was stored at 4℃ for 90 d,it could not make the preadipocytes develop soundly,even when L-Gln was added. Once more,M199 and FCS (fetal calf serum)could not be stored at 4℃ beyond 90 d if they were opened. The above shows that suitable supplement of L-Gln could make preadipocytes approach better states of proliferation and differention.
