不同应激对HeLa细胞增殖、凋亡及热休克蛋白70表达的影响 被引量:5

摘要 背景与目的:研究热应激、地塞米松(Dexamethasone,Dex)、吐温80(Tween_80)、氨茶碱、乙二胺四乙酸(Ethylenediaminetetra_aceticacid,EDTA)、Ca2+和双氧水应激对HeLa细胞增殖、凋亡及热休克蛋白70(Heatshockprotein70,HSP70)表达的效应。材料与方法:应用噻唑蓝法[3_(4,5_dimethylthiazol_2_yl)_2,5_diphenyltetrazolium,MTT]测定细胞增殖;将收集的HeLa细胞制备成玻片和硝酸纤维素膜(Nitrocellulosemembrane,NCM)两种标本,应用免疫细胞化学、斑点印迹技术检测HSP70;提取细胞DNA,应用琼脂糖凝胶电泳检测细胞凋亡。结果:HeLa细胞经不同应激处理后,细胞增殖均受到抑制;除EDTA处理组未出现凋亡外,其余组均见DNA降解,形成梯形条带;各处理组HSP70表达较对照组有所提高,其中双氧水、热处理和氨茶碱处理组与对照组相比差异有统计学意义,Dex和Tween_80处理组与对照组相比差异有统计学意义,EDTA和Ca2+处理组HSP70表达有升高趋势,但差异无统计学意义。结论:不同应激可提高HeLa细胞的HSP70表达,并引起细胞不同程度的凋亡。 BACKGROUND & AIM: To investigate the effects of different stress factors on HeLa cell proliferation, apoptosis and heat shock protein 70(HSP70) expression. MATERIAL AND METHODS: Cell culture technique, 3-(4, 5-dimethyhhiazol-2-yl) -2, 5-diphenyl tetrazolium(MTF) flow cytometry, immunocytochemistry, western dot. RESULTS: The present study showed that cell apoptosis appeared in all experiment group; there was a tendence to increase HSP70 expression treated with Ethylenedianline tetra-acetic acid (EDTA) and Ca^2+; The protein level of HSP70 was elevated significantly in the groups of H2O2, heating and aminophylline (P〈0.01) than control group; the protein level of HSP70 was higher (P〈0.05) than control group in the groups of dexanlthasone and tween-80. CONCLUSION: The results suggested that the expression of HSP70 in HeLa cells could be elevated and apoptosis could be induced by stress factors.
