摘要
目的观察连续给予穿心莲内酯亚硫酸氢钠加成物后,由于积蓄而对机体产生的毒性反应及严重程度,提供中毒靶器官及其损害的可逆性,确定无毒反应剂量。方法大鼠连续4周腹腔给予穿心莲内酯亚硫酸氢钠加成物250、500、1000mg.kg-1,观察动物的一般状况,体重增长及血液、生化指标并解剖进行病理检查。结果穿心莲内酯亚硫酸氢钠加成物250mg.kg-1给药4周后大鼠一般行为、体重增长、尿常规、血液学、血液生化学、脏器重量、脏器系数及病理学检查均未见显著性变化。500mg.kg-1给药结束后BUN、CRE有升高趋势。1000mg.kg-1给药后动物出现扭体、消瘦、体重减轻等症状;给药结束后BUN、CRE升高,组织病理学检查3/10动物出现肾小管出血,小灶性水肿。以上各症异常指标在停药后均可恢复。结论连续腹腔注射4周,安全用药剂量为250mg.kg-1.d-1。
Objective To observe the toxic response and the serious degree,the target organ and the toxic reversibility and to determine the non-toxic dosage,the chronic toxicity study on the adduct of andrographolide with sodium hydrosulfite in rats was carried out.Methods The adduct of andrographolide with sodium hydrosulfite was administered to the rats by i,p.in doses of 250,500 and 1000 mg· kg^-1·d^- 1 within 4 weeks. Record the Clinical signs and body weight growth,and examine the haematology, clinical chemistry,gross necropsy and histopathology. Results In the group of 1000 mg· kg^-1, the below findings were observed： suppression in body weight gain,the level of BUN and CRE devate.3/10 animals have the renal tubule hemorrhage and special mess hall dropsy. In the group of 500 mg· kg^- 1, the level of BUN and CRE has the tendency to rise. All of the changes above were disappeared after the recovery period. No change were seen in the group of 250mg· kg^- 1. Conelnsion It was concluded that the safe dose of the adduct of andrographolide with sodium hydrosulfite is 250mg· kg^-1· d·-1 in this study.
