摘要
目的改进采用人骨髓体外培养人骨髓基质细胞(hBMSCs),进行体外传代扩增,并探索体外诱导分化为神经元样细胞的方法.方法抽取人骨髓血,淋巴细胞分离液分离,DMEM/15%人血清培养传代,至第3~5代时加入巯基乙醇(BME),观察hBMSCs分化为神经元样细胞的形态学变化.结果该方法hBMSCs在体外可以实现数目扩增,在特定诱导剂的作用下向神经元样细胞分化.结论采用少量骨髓样本,既可培养获得充足的细胞量,可以满足定向诱导和细胞移植的需要.
Objective To investigate a new methods of hBMSCs differentiating into neurons, human bone marrow was isolated and cultured in vitro. Methods hBMSCs were separated with lymphocytic cell separating medium, hBMSCs were cultured and expanded in vitro. Then at passage 3 hBMSCs were induced with β-mercaptoethanol (BME). Neuron morphology was observed. Results hBMSCs were able to expand in vitro and BME were able to induce cells into neuron. Conelusion A few of hBMSCs can obtain sufficiently cells for induction and cells transplantation.
