多脏器功能障碍综合征患者外周血单个核细胞凋亡的意义研究 被引量:3

摘要 目的观察多脏器功能障碍综合征(MODS)患者外周血单个核细胞(PBMCs)的细胞凋亡。方法采用流式细胞仪法、电子显微镜、荧光显微镜测定25例MODS患者PBMCs的细胞凋亡,并与18例健康志愿者比较。结果电镜、吖啶橙—溴化乙锭染色荧光显微镜均观察到MODS患者PBMCs出现细胞凋亡的典型形态学改变;MODS组和健康对照组的PBMCs凋亡率分别为(25.4±9.21)%、(15.9±6.75)%(P<0.01),凋亡细胞数分别为1.040±0.096、0.235±0.028个/HPF(P<0.05)。结论MODS患者PBMCs细胞凋亡异常增高。 Objective To investigate PBMCs apoptosis in the patients with MODS. Methods Flow cytometry assays, electron microscope, acfiding orange- ethidium bromide staining and fluorescence microscope , DNA agorese gel electmphoresis were used to identify and quantify apoptosis of PBMCs in 25 patients with MODS. 18 healthy volunteers were selected as control group.Results Typical morphological features of apoptotic PBMCs in the patients were observed under electron microscope and fluorescence microscope by using AO - EB staining and DNA agorese gel electrophoresis .The apoptosis rate of PBMCs in MODS group was (25.4 ± 9.21 ) %, which was higher than that in control group ( 15.9 ± 6.75) % ( P 〈 0.01 ). The apoptotic cells were ( 1.040 ± 0.096) piece/HPF in MODS group, which was higher than that in control group (0.235 ± 0.028) piece/HPF (P 〈 0.05) by counting under AO - EB staining and fluorescence microscope. Conclusions PBMCs apoptosis in the patients with multiple organ dysfunction syndrome are increased abnormally.
