摘要
目的:对Ca2+在兔不全梗阻性不稳定膀胱逼尿肌细胞中的变化进行研究。方法:成年同龄雄性新西兰白兔30只,其中15只为膀胱出口不全梗阻8周,尿流动力学证实为不稳定膀胱者为实验组,15只仅手术游离兔膀胱颈而不做结扎梗阻为对照组。模型成熟后,采用急性酶法分离及传代培养的方法获得单个膀胱平滑肌细胞。对培养的第一代细胞采用激光共聚焦显微镜观察模型膀胱平滑肌细胞静态Ca2+浓度。结果:静息状态下不全梗阻性不稳定膀胱平滑肌细胞内Ca2+浓度明显增高(Ca2+超负荷)。结论:膀胱逼尿肌细胞Ca2+病理性改变是出现不稳定膀胱的重要因素之一。
Objective :To study Ca^2+ changes in detrusor cell of the unstable bladder of rabbit's partial bladder outlet obstruction. Methods: Thirty male New Zealand White rabbits were divided into 2 groups randomly. In the experiment group the unstable bladder were confirmed by Urodynamics and the sham operated age-matched rabbits acted as the control group eight weeks after operetion, bladder smooth muscle ceils were isolated by collagenase digestion. Intracellular Ca^2+ levels in detrusor cell were observed by confocal laser scanning microscope (CLSM). Results: Intracellular Ca^2+ concentration significantly increased in the experiment group (Ca^2+ overload). Conclusion:It was a important pathogeny for the unstable bladder of partial bladder outlet obstruction that pathological changes of Ca^2+ with the unstable bladder.
