摘要
目的探讨米诺地尔对增生性瘢痕成纤维细胞增殖和细胞结构的影响。方法体外原代培养增生性瘢痕成纤维细胞,将0.125~4.0mmol/L的米诺地尔分别作用增生性瘢痕成纤维细胞12h,24h,48h后,采用MTT、流式细胞仪、透射电镜方法检测细胞的增殖活性及其超微结构的变化。结果MTT发现0.25~4.0mmol/L浓度的米诺地尔对增生性瘢痕成纤维细胞的增殖有抑制作用。用药48h后,米诺地尔组细胞的体积变小,核变小,核仁变小,胞内有许多溶酶体出现,未见到明显的细胞凋亡;用药48h后流式细胞仪检测发现,细胞周期中用药组比对照组S期细胞减少10.1%,G1期增加了10.4%。结论米诺地尔对增生性瘢痕成纤维细胞的增殖有抑制作用。其作用机制:米诺地尔抑制胶原的合成,另外可能增加K+外流,使细胞膜超极化,减少Ca2+内流使细胞的Ca2+浓度下降及降低细胞内的自由基生成来抑制瘢痕成纤维细胞的增殖。
Objective To study the effect of minoxidil on the hypertrophic sear derived fibroblasts. Methods The fibroblasts were derived from the cultured hypertrophic scar tissue. After the cells were treated with different concentration of minoxidil, the proliferation of hypertrophir scar fibroblasts were measured with eolormetrie MTT, flow cytometry, anti transmission electron mieroseope. Results Minoxidil (0.25, 0.5, 1,0, 2.0, 4.0mmol/L) could inhibit the proliferation of hypertrophic scar fibroblasts (P 〈 0.01 ). Compared with the control groups, the distribution in cells was shown that percentage of S phage decreased ,while the percentage of G1 phage cell increased. Compared with the control groups,transmission electron microscope (TEM) shown that the volume of cell decreased,and nueleus decreased.lysosome increased too. Conclusion Minoxidil eould inhibit the proliferation of hypertrophic scar fibroblasts.
