摘要
目的探讨RNA干扰bcl-2基因对人卵巢癌细胞体外增殖及凋亡的调控作用.方法选用人卵巢癌细胞进行体外培养,以siRNA分别处理SKOV3细胞24~72h后,MTT法检测细胞增殖,SABC免疫组织化学法检测细胞内bcl-2、bcl-xl、bax、caspase-3蛋白表达水平.结果 RNA干扰bcl-2基因对SKOV3细胞增殖有抑制作用.bcl-2表达下调,Caspase-3蛋白表达水平上调.结论 RNA干扰bcl-2对人卵巢癌细胞的增殖具有明显的抑制作用.下调bcl-2的表达,上调Caspase-3蛋白表达水平.特异性地降解抗凋亡基因bcl-2的mRNA是其作用机制.
Objective To investigate the regulatory effect of RNA interference on proliferation and apoptosis on human ovary carcinoma cells SKOV3 in vitro. Methods Human ovary carcinoma cell line SKOV3 was cultured in vitro. SKOV3 cell were treated with siRNA for 24h to 72h and the growth inhibition rates of SKOV3 cells were measured with MTT method. In addition,the expression of bcl-2,bcl-xl,bax and caspase-3 protein in SKOV3 cell was observed by SABC immunohistochemistry. Results RNA interference inhibited the proliferation of SKOV3 cell on a time depending manner . The bcl-2 expression was decreased while caspase-3 expression was increased. Conclusion RNA interference could significantly inhibit the growth of SKOV3 cells, inducing apoptosis through up regulating expression of caspase-3 and down regulating expression of Bcl-2 . The results demonstrate that specific degradation of the mRNA of the gene is mechanism of RNA interference.
