摘要
目的检测幽门螺杆菌(Hp)感染患儿胃黏膜中白细胞介素-12(IL-12)及IL-12信使核糖核酸(mRNA)变化,从蛋白和分子水平探讨Hp对细胞因子IL-12的影响,以及IL-12在Hp相关性胃炎中的作用。方法胃镜下取Hp患儿胃窦黏膜活检标本,用ELISA法和RT-PCR定量法测定胃窦黏膜IL-12及IL-12mRNA表达量。结果Hp阳性者胃黏膜中IL-12和IL-12mRNA分别为(66.42±15.15)ng/g,(59.21±15.03)%;Hp阴性者胃黏膜IL-12和IL-12 mRNA分别为(22.22±8.79)ng/g,(17.94±7.39)%,差异有显著意义(P<0.01)。结论Hp阳性胃炎患儿胃黏膜IL-12及IL-12 mRNA表达量均增高,提示IL-12在Hp相关性胃部疾病致病过程中起关键作用。
Objective To determine the changes of interleukin- 12（IL- 12） and IL- 12 nLRNA in gastric mucosa of children with helicohacer pylori （Hp） infection,and to study the effects of Hp infection on the expression levels of IL - 12 and IL- 12 mRNA, and to evaluate its possible roles in the pathogenesis of gastric mucosal inflammation in Hp related gastroduodenal diseases. Methods Biopsy specimens were taken from the antral mucosa on endoscopy in patients with or without Hp infection, which were diagnosed by urease test and （iierusa staining. The expression levels of IL- 12 and IL- 12 mRNA in gastric mucosa were determined by ELISA and RT- PCR. Results Inflamnation of gastric antral mucosa was more severe in Hp- positive mucosa . The expression levels of IL- 12 and IL- 12 mRNA m Hp- positive mucosa were （66.42 ± 15.15） ng/g and （59.21±15.03）% ,which were more than those in Hp- negative（22. 22 ±8. 79） ng/g and （17.94±7.39）% （p〈0.01）. Conclusions The expression levels of IL- 12 and IL- 12 mRNA are higher in Hp-positive gastritis. IL- 12 may play a pivotal role in the Hp associated gastric disease.
