ANALYTICAL EVALUATION OF FLUORESCENCE POLARIZATION IMMUNOASSAY FOR PLASMA HOMOCYSTEINE

摘要 ①目的对荧光偏振免疫法(FPIA)测定血浆同型半胱氨酸(HCY)的方法学进行初步评价.②方法应用FPIA法测定HCY的精密度、标准曲线、回收率并与高效液相色谱法(HPLC)测定HCY的结果进行相关性分析.对80例健康体检者的血浆HCY浓度用FPIA法进行了测定,以确定其参考值.③结果FPIA法测定HCY的精密度(批内CV为1.16%,批间CV为3.12%)较高;HCY浓度在0~50.0 μmol/L之间,线性关系良好;回收率为103.0%.FPIA法和HPLC法测定HCY结果呈高度正相关(r=0.985,P<0.01).健康体检者HCY参考值范围:男性为4.96~15.83 μmol/L,女性为5.02~14.15 μmol/L.④结论FPIA法测定血浆HCY具有较高的精密度,结果准确可靠,适合临床实验室推广应用. Objective To evaluate the detection of plasma homocysteine (HCY) with fluorescence polarization immunoassay (FPIA) technology. Methods The precision, calibration curve, recovery of the FPIA assay were determined. In order to determine reference value of HCY concentration, plasmas were determined in 80 healthy individuals. Results The FPIA method revealed good precision (CV 1.16% ,3.12%) and recovery (103.0%). Linear regression analysis showed a very good positive correlation between FPIA and HPLC methods (r=0. 985,P〈0. 01). Reference range of healthy subjects was; male 4. 96- 15.83 μmol/L, female 5.02- 14. 15 μmol/L. Conclusion The FPIA method is precise, reliable and suitable for determinations of plasma HCY concentration in a clinical laboratory.
