摘要
目的探讨人嗅鞘细胞的免疫学特征。方法应用流式细胞仪分析人嗅鞘细胞的相应标记抗原的阳性表达率,用单向混合淋巴细胞反应测定人嗅鞘细胞的组织相容性和免疫耐受性,并与骨髓间充质干细胞对比。结果人嗅鞘细胞与骨髓间充质干细胞表面与移植免疫排斥发生密切相关的标志:HLADR、B71(CD80)、B72(CD86)、CD40和CD40L均为阴性。单向混合淋巴细胞反应显示人嗅鞘细胞、骨髓间充质干细胞均对混合淋巴细胞反应无明显的刺激作用。结论人嗅鞘细胞可能是一种免疫缺陷细胞,在体外有诱导细胞移植免疫耐受性的作用。
Objective To investigate the immunologic characters of human olfactory ensheathing cells (OECs). Methods Human OECs were obtained from olfactory bulbs and MSCs were isolated from human bone marrow aspiration, stained with fluorescein isothiocyanate ( FITC)-conjugated or phycoerythrin (PE)-conjugated monoclonal antibodies, analyzed by using a FACScan laser flow cytometry system. Lyrnphocytes were mixed at a 1:1 ratios with OECs/MSCs in MLR. The proliferation of lyrnphocytes was measured on the day 6 as mean 3H-TdR uptake in counts per min. Results Both OECs and MSCs were negative for HLA-DR, CD80 (B7-1 ), CD86(B7-2 ), CD40, CS40L, histocompatibility antigens related to GVHD or tolerogenic activities. OECs and MSCs could wealdy stimulate the proliferation of lyrnphocytes in MLR compared with the control MLR, but Student t test showed no significant difference (P〉0.05). Conclusion Human OECs are not significantly immunogenic.
