摘要 目的研究17β雌二醇(E2)对大鼠血管平滑肌细胞(VSMC)凋亡的影响及其可能机制。方法应用流式细胞术检测不同浓度E2(0-100nmol/L)在有或无血清状态下对传代VSMC凋亡及其相关蛋白Bcl-2和Bax表达的影响;并采用免疫荧光染色和共聚焦激光扫描荧光显微镜术对Bax的分布进行定位。结果在有或无血清条件下,当浓度达到10nmol/L以上时,E2引起VSMCBax蛋白表达上升及Bax/Bcl-2比值的改变,并促进VSMC凋亡。这一效应有随E2剂量和作用时间增高的趋势。结论E2可通过上调Bax蛋白表达而诱导VSMC凋亡。 Objective To investigate the effect of 17β-estradiol (E2) on the apoptosis of vascular smooth muscle cells (VSMC) and the possible underlying mechanisms. Methods The rate of apoptosis and the expressions of Bcl-2 and Bax proteins were examined by flow-cytometry in VSMC cultured in different concentrations (0--100nmol/L) of E2 with or without serum. The distribution of tAax in VSMC was detected by using immunofluorescent staining and confocal laser scanning fluorescence microscopy. Results At the concentrations of 10 nmol/L or higher, E2 .induced an increased expression of Bax protein with changes in the Bax/Bcl-2 ratio. This was accompanied by an increase in the apoptotic rate of VSMC cultured with or without serum. Conclusion E2 induces the apoptosis of VSMC by up-regulating Bax protein expression.
