摘要 目的观察缺氧对培养肝细胞极性的影响。方法利用三明治构型培养成鼠肝细胞,观察缺氧后肝细胞形态、特异性膜区域蛋白分布以及胞质钙离子的变化,以确定对细胞极性的影响。结果缺氧后,肝细胞变得扁平,分化良好的小管样结构消失;免疫组织化学证明肝细胞膜小管区域蛋白失去特异性分布;激光共聚焦显微镜显示肝细胞胞质钙离子荧光强度增强。结论缺氧可导致肝细胞极性丧失,从而造成肝细胞损伤,本研究可为在细胞水平研究肝脏缺血性再灌注损伤的发病机制提供一个体外模型。 Objective To investigate the effects of hypoxia on polarity of hepatocytes cultured in vitro. Methods Sandwich- structural hepatocytes were used to study the effects of hypoxia on polarity of cultured hepatocytes. Expression of specific plasma protein, detensity of cytosolic Ca^2+ and the morphology was observed before and after hypoxia. Results Both bile canaliculia-like structures and the distribution of specific plasma membrane protein were damaged in hypoxia treated hepatocytes. The detensity of cytosolic Ca^2+ was increased in hypoxic hepatocytes. Conclusion Hypoxia induces significant disturbance of cell polarization in hepatocytes.
