摘要
探讨剪切力对脐静脉内皮细胞内游离钙的影响,利用流室装置,通过精密蠕动泵提供剪切力,选择5×10-5N/cm2(5dyne/cm2)和10×10-5N/cm2(10dyne/cm2)两种剪切力作用体外培养的脐静脉内皮细胞,作用不同时间,用荧光染色法来观察细胞内游离钙的变化。结果细胞在受到剪切力作用下,其胞内钙离子均有显著增加,且10×10-5N/cm2(10dyne/cm2)组要高于5×10-5N/cm2(5dyne/cm2)组,两实验组与对照组相比,两实验组之间相比在统计学上均具有显著性差异。说明内皮细胞受到剪切力作用后其细胞信号传导机制可能与细胞内钙的变化有关。
The objective was to study the effects of dissociative Ca^2+ in vascular endothelial cells by different shear stresses. Utilized parallel plate flow chamber and pump to control the value (5 dyne/cm^2, 10 dyne/cm^2) and duration (15 min, 30 min) of shear stress in vascular endothelial cells. The concentration of Ca^2+ in HUVEC was measured by staining the cells with Flou-3AM. It was found that the concentrations of intracellular Ca^2+ was higher than that of the control groups under 5 dyne/cm^2 and 10 dyne/cm^2 shear stress. However, the levels of Ca^2+ in the groups of 10×10^-5 N/cm^2 (10 dyne/cm^2) was higher than that of in 5×10^-5 N/cm^2 (5 dyne/cm^2). The result shows that cells signal mechanotransduction induced by shear stress might have related to the changes of intracellular Ca^2+.
