摘要
目的:观察承气生血方对小鼠Lewis肺癌肿瘤细胞的抑制作用。方法:采用动物移植性肿瘤实验法,腋皮下接种Lewis肺癌,随机分组,口服给药后计算平均抑瘤率。以DNA探针进行肿瘤细胞荧光标记,荧光显微镜观察凋亡细胞形态,并以流式细胞术分析肿瘤细胞周期变化及凋亡百分率。结果:西药对照环磷酰胺组肿瘤抑制率达89.2%(P<0.01);承气生血方1.2g·kg-1d-1组肿瘤抑制率36.1%(P<0.01)。荧光显微镜观察,环磷酰胺组偶见凋亡细胞;承气生血方组可见大量凋亡细胞及坏死细胞。流式细胞分析,环磷酰胺组肿瘤细胞比例为25.89%,肿瘤细胞凋亡率4.48%,与模型组比较具有显著性差异(P<0.01);承气生血方1.2g·kg-1d-1组肿瘤细胞的比例虽然占49.07%,与模型组比较仍具有显著性差异(P<0.05),肿瘤细胞凋亡率平均达到29.94%,与模型组比较具有显著性差异(P<0.01)。结论:西药环磷酰胺可直接杀伤小鼠Lewis肺癌细胞,具有强大的细胞毒作用。承气生血方作用较缓和,同样具有抗小鼠Lewis肺癌作用,并可通过诱导肿瘤细胞凋亡抑制肿瘤细胞生长。
Objective: To study the effect of Chengqi Shengxue Formular on inhibiting mouse Lewis Lung Carcinoma tumor growth. Methods: A piece of Lewis Lung Carcinoma was transplanted into the subcutaneous space on the armpit of each C57BL mouse. Mice were randomized into several groups. After orally administered, the mean weight of the subcutaneous tumors was examined, then the average tumor control rate was analysed. Tumor cells were labeled by DNA fluorescence probe, then the apoptosis of tumor cells were observed by immunofluorescence microscope. Further, the cell cvcle and apoptotic cell rate were calculated flow cytometer (FCM) . Results: In the group treated by Cyclophosphamide, the tumor control rate was 89.2 % (P〈0.01) ;compared to the tumor model group. In the group treated by Chengqi Shengxue Formular 1.2g·kg^-1d^-1, the tumor control rate was 36.1% (P〈0.01); compared to the tumor model group. By immunofluorescence microscope, many apoptotic and necrosis cells were observed in the group treated by Chengqi Shengxue Formular while only a few apoptotic cells were observed in the group treated by Cyclophosphamide. In Cyclophosphamide group and Chengqi Shengxue Formular group, the percentage of tumor cells was released to 25.89% ( P〈0.01 ) and 49.07%( P〈0.05) respectively. At the same time, in these two groups,the apoptotic tumor cell rate was 4.48% (P〈0.01); compared to the tumor model group and 29.94%(P〈0.01) respectively. Conclusion: These results suggest that Chengqi Shengxue Formular could inhibit the growth of mouse Lewis Lung Carcinoma, and its mechanism may be related with its action of inducing the apoptosis of tumor cells. Cyclophosphamide can kill mouse Lewis lung carcinoma cells in a direct way due to its cytotoxinic effect.
