摘要
目的观察牛磺酸对兔角膜基质细胞增殖的影响。方法将体外培养的原代兔角膜基质细胞传至2-3 代用于实验,分别加入25、50、100、200、400mmol/L的牛磺酸,用药后2、4、6、8天倒置显微镜下加台盼蓝染色活细胞计数,计算细胞抑制率,并于2、4、6天进行MTT方法检测。结果细胞计数和MTT法均显示牛磺酸浓度≥100 mmol/L时,作用2天、4天和6天对兔角膜基质细胞增殖均有明显的抑制作用,其抑制作用呈剂量依赖性。结论牛磺酸具有抑制兔角膜基质细胞增殖的作用,该研究结果为防治PRK和LASEK术后角膜上皮下混浊(haze)形成提供了客观依据。
Objective To study the effect of taurine on the proliferation of cultured rabbit corneal keratocytes. Methods Two or three generation of cultured rabbit corneal keratocytes were added with taurine (25,50, 100,200,400mmol/L). Cells were counted under reverse microscopy and MTT technique was employed on 2, 4,6 days posttreatment with taurine. The effect of taurine on cultured rabbit keratocytes proliferation was evaluated. Results Both MTT and cell count showed that taurine inhibited the proliferation of cultured rabbit keratocytes when the concentration of taurine was greater than 100 mmol/L. The inhibition of proliferation was concentration dependent. Conclusion Taurine inhibits the proliferation of cultured rabbit corneal keratocytes. These results provide a preliminary theoretical basis in preventing haze after PRK and LASEK.
