摘要
卡那霉素链霉菌温和性噬菌体SKJ1DNA上有2个AvaⅠ和SmaⅠ酶切位点,8个SalⅠ位点,7个EcoRⅠ位点及12个BamHⅠ位点。经酶切后用1%琼脂糖凝胶电泳测定SKJ1DNA大小为60.7kb.用单酶切、双酶切及片段酶切分析,得到SKJ1DNA的AvaⅠ、SmaⅠ、EcoRⅠ、SalⅠ的酶切图谱及BamHⅠ的部份酶切图谱。证明AvaⅠ和SmaⅠ在SKJ1DNA上具有相同酶切位点。并经过酶切结果分析和图谱构建,SKJ1DNA可能为1个环状分子。
Actinophage SKJ1 DNA has 2 cut sites for AvaⅠand SmaⅠ,8 for SalⅠ,7 for EcoRⅠand 12 for BamHⅠ.The size of SKJ1 DNA was estimated to be about 60.7 kbby means of electrophoresis on 1%agarose gel digested with various enzymes. With single,double enzyme digestiom and fragment isolation,the restriction map of SKJ1 DNA for AvaⅠ,SmaⅠ, EcoRⅠ,SalⅠand a partial restriction map for BamH Ⅰ have been constructed.Ithas shown that AvaⅠaidSma Ⅰ have the same cut sites on SKJ1 DNA. Study on theenzyme digestion and the construction of the restriction map suggested SKJ1 DNA to be acircular molecule.
