摘要
茯苓素在小鼠体内诱生肿瘤坏死因子的机制在于增强TNF基因的转录。实验结果表明:莅菩素以27~81mg/kg剂量刺激小鼠巨噬细胞,可得到较高效率的mRNA表达,超过此剂量可由于药物的毒性作用而造成基因转录水平的下降。mRNA的杂交结果(SlotBlotting)可通过Tubulin探针的杂交而得到验证。
The mechanism of stimulation of TNF production by macrophages causedby poriatin was related to enhancement of transcription of TNF gene.Experiments showedthat higher effcient expression of TNF mRNA can be obtained with poriatin at doses of 27~81mg /kg,Over the optimum dosage,the level of transcription of TNF mRNA was loweredand it might be due to the toxicity of the drug.The result of slot blotting was identified by rehybridization of α-tubulin probe.
