酸枣微繁技术的研究 被引量:17

摘要 酸枣微繁研究,成功地解决了消毒、繁殖、生根及移栽等关键技术问题。二次升汞灭菌法Ⅱ,材料污染率仅18.1%,比一次升汞灭菌法1效果提高4.2倍。不同发育阶段适宜培养基组成(mg/L):促进萌芽生长培养基有MS+BA0.5mg/L+NAA0.5mg/L和MS+BA3.0mg/L+IBA1.0mg/L二种;继代繁殖培养基:MS+BA0.1—0.5mg/L、+NAA0.1mg/L培养40天,平均苗高4.64—3.82cm,芽繁殖倍数为7.6—6.3;生根培养基为1/2MS+IBA1.0mg/L+IAA1.0mg/L,最高生根率为100%。试管苗移栽成活率97.9%,定植成活率100%。近三年研究一个简易、快速、实用酸枣工厂化育苗生产程序已基本建成。 This experiment has solved basically some key problems of the technology of the clonal porp-agation and the factory culture seedling of wild jujube :(1)The new way of the high efficient dis-infecion that is HgCl_2 disinfecion twice,the frequency of the meterial polluted was only 18.1%,the effect of this way has rasied more four times than the way of Hg Cl_2 disinfecion one time;(2)The axillary bud and grow the medium: MS+BA0.5mg/L+NAA 0.5mg/L; MS +BA3. 0mg/L+IBA1. 0mg/L;(3)The medium of the succssion porpagate:MS+BA0.1 0. 5mg/L+NAA0.1mg/L. No.of proliferation times of buds was 7.6 6. 3,after 40 days;(4)The medi-um of growing root:1/2MS+IBA1.0mg/L+IAA1. 0mg/L,The concentration of sucrose of ev-ery medium is 2%.The lilving rate of the transplantation of the tube plant was 97.9%.The liv-ing rate of field planting was 100%.A set simple practial and quick production process of factory culture seedling of tuble。seedling has proliminary established.
