锌对大鼠急性缺血心肌溶酶体膜的影响

摘要 本实验结扎大鼠冠状动脉,造成急性心肌梗塞模型,观察锌对缺血心肌溶酶体酶的影响。结果表明,心肌缺血1h后,缺血区心肌溶酶体酸性磷酸酶、组织蛋白酶D游离酶(F)及游离酶/总酶比值(F/T)明显升高,GSH-Px活性显著降低,脂质过氧化产物丙二醛(MDA)含量明显升高。而锌预处理大鼠心肌急性缺血后,F及F/T均显著下降,MDA含量明显降低,但GSH-Px活性无明显改变。故锌可能通过GSH-Px以外的途径,增加溶酶体膜的稳定性,减轻心肌缺血性损伤。 In this paper the coronary artery of rat was ligated to make the model of myocardial infarction, the effect of zinc on ischemic myocardial lysosomal enzyme was observed. The results indicated that the activities (F) of lysosomal free acid phosphatase (ACP), cathepsin D (CD)and their total enzyme activity ratios (F/T) were all increased significantly in ischemic area 1h after ischemia. Simutaneously, the GSH-Px activity decreased significantly, the content of lipid peroxidation product MDA was significantly increased. In the rat pretreated with zinc, the F and F/T decreased significantly in ischemic area after acute myocardial ischemia, the MDA content decreased obviously, the GSH-Px activities had no significant change. Zinc could stabilize the ischemic myocardial lysosome and protect ishemic myocardium from ischemia injury by other mechanism rather than raising GSH-Px activity.
