过氧化物酶在丝素膜中的固定化及其活性研究 被引量:2

摘要 家蚕Bombyxmori丝素蛋白经中性盐溴化锂处理后成为水溶性丝素,然后制成固定化过氧化物酶(POD)的丝素膜。经分光光度法分析表明这种固定化POD丝素膜的最适pH值范围较游离酶宽,为pH7.0~8.0;酶促反应温度在50℃其活性最大。而且,此固定化酶膜的热稳定性良好,在60℃保温15分钟,其活性几乎未受影响,而游离酶失活40%。因此,POD对pH和热的稳定性在丝素蛋白中固定后得到了改善,蚕丝丝素是一种良好的固定化酶的生物材料。 The aqueous solution of the silk fibroin from silkworm Bombyx mori cocoons was obtained by treatment of the neutral salt(LiBr). Peroxidase (POD) was immobilized in the silk fibroin membrane by immersion in 80% methanol aqueous solution.Optimum pH range of the activity of the POD-immobilized fibroin membrane.being at pH7. 0-8. 0,was widened in comparison with the case of free enzyme. The maximal activity for the immobilized enzyme was at reaction temperature 50℃ (pH7. 0),and its thermal stability was good since its activity was scarcely lost althrough the free POD lost about 40% of the original activity,when being incubated at 60℃ for 15min. Thus the stabilities of the enzyme VS. pH and heat were much improved by the immobilization in silk fibroin. Silk fobroin is a good biomaterial for immobilized enzyme.
