运用多谱勒超声心动图对小鼠胚胎心脏血流动力学的研究

摘要 对210个C_(57)BL/6J实验小鼠胚胎心脏进行了多谱勒超声心动图研究。胎龄从10d至19d。发现随胎龄的增加,心率、心室充盈时间显著增加;心脏等容收缩时间至17d降至0;心室射血时间和等容舒张时间无明显变化。这与从人类胚胎心脏研究所得结果很相似小鼠作为对人类胚胎心脏发育研究的合适模型,为此提供了完整的正常对照数据,尤其为研究转基因的动物模型开创了非创伤性监测胚胎早期心脏血流动力学的途径。 To analyze mammalian embryonic heart hemodynamics, Doppler echocardio-grams are performed on C57BL/6J mouse embryos from day 10D(10) through D19 of gestation. Heart rate increases with increasing gestational age. The time period of isovolemic contraction time decreases during development down to 0 on D17, while the total filling time increases a similar amount. Ejection time and isovolemic relaxation time have no significant change. These echocardiographic patterns are similar to those observed for human embryos during fetal life. It indicates that the mouse embryo is an appropriate animal model to analyze normal and abnormal mammalian heart development and function. The presented echocardiographic data can serve as a baseline for further study of abnormal heart development. This noninvasive technique may be invaluable to monitor especially those transgenie embryos where heart defects are highly expected.
