摘要
萱草(Hemerocallis fulva L.)单核后期花粉,经花蕾低温预处理后由酶法分离出原生质体。在含多种附加物的 K_3培养基上,部分花粉原生质体启动细胞分裂,形成了多细胞团与原胚。显微观察表明,第一次胚胎发生分裂有均等与非均等分裂二种方式,以后进行有组织或无组织生长,导致了多细胞团与原胚并存以及结构上的多样性。本文是第一篇关于花粉原生质体培养启动胚胎发生的报道。
Pollen protoplasts were isolated from the cold-pretreated flower buds containing uninuc- leate microspores and were inoculated in K_3 basic medium supplemented with various additives- A part of the pollen protoplasts were triggered to embryogenic divisions and further to for- marion of proembryos and clusters.Microscopical observations showed that the first cell division might be cqual or unequal and the subequent growth might be organized or unorga- nized.This is the first example of pollen protoplast culture in which a sporophytic,or embry- ogenic,pathway is triggered
