摘要
目的研究烟曲霉菌菌丝片段和孢子对气道上皮细胞的作用。方法烟曲霉菌菌丝片段和照射灭活的孢子分别与气道上皮细胞系A549共培养,酶联免疫吸附实验(ELISA)检测A549细胞释放到培养液内的白细胞介素(IL)-6和IL-8。结果菌丝片段刺激A549细胞产生有限的IL-6、IL-8;孢子抑制A549释放IL-6。结论烟曲霉菌菌丝片段和孢子被吸入气道后,致少量细胞因子甚至抑制细胞因子产生,不能使足够的炎症细胞向局部趋化,逃避吞噬,长期存活、芽生、释放大量过敏原。
Objective To investigate the interaction of aspergillus fumigatusand and airway epithelial cellline A549 in vitro. Methods The strain of aspergillus fumigatus was isolated from the sputum of a patient who died of aspergilloma in Groningen University Hospital. The mycelium and spores of spergillus fumigatus were treated with heat and irradiation and then used to cocultul'e with A549 cells. After 24 h or 48 h the supernatant was harvested and the levels of IL-6 and IL-8 secreted by A549 cells were measured using ELISA. The cytospins were prepared and stained with MGG. Results After cocuhure with irradiated spores and mycelium A549 cell had no morphology changes. After washing the fragments of mycelium, heated mycelium were firmly binding to the surface of A549 cells and so did the irradiated spores. In addition, A549 cells engulfed some of spores. The positive control of TNF-α led to tense release of IL-6 and IL-8 by A549 cells while the fragments of mycelium induced limited produc- tion of IL-6 and IL-8 by A549. Notably, the spores of aspergillus fumigatus even inhibited A549 cell to produce IL-6. Conclusion The low levels of cytokines secreted by airway epithelial cells after the firm binding of either mycelium or spores may lead to insufficient recruitment of inflammatory cells to the infected site, which may result in escape of detection by immune system.
