降钙素基因相关肽对新生鼠HIE脑组织内皮素及脑细胞内钙的影响 被引量:2

Influence of calcium gene-related peptide on endothelin and free calcium concentration in brain cells of hypoxic-ischemic neonatal SD rats.

摘要 【目的】研究降钙素基因相关肽(calciumgene-relatedpeptide,CGRP)对新生鼠缺氧缺血性脑损伤(hypoxic-ischemicencephalopathy,HIE)脑组织内皮素(endothelin,ET)及脑细胞内游离钙离子(intracellularfreecal-cium,IFCa)的影响。【方法】将45只7日龄SD(Spraque-Dawley)大鼠平均分成3组:①CGRP治疗组于HIE模型后即给予CGRP3μg/(kg.d)加生理盐水(NS)2ml稀释后,腹腔内注射,共3d;②NS组于HIE模型后即给予NS2ml/d,腹腔内注射,共3d;③正常对照组(假手术组)。各组于3d后断头取脑,采用放射免疫法检测脑组织ET含量,应用钙荧光指示剂Fura-ZAM法测定IFCa。【结果】NS组ET及IFCa含量[(93.41±12.73)ng/L及(129.47±17.21)nmol/L]明显高于正常对照组[(57.58±6.31)ng/L及(93.72±24.16)nmol/L]和CGRP治疗组[(61.68±6.83)ng/L及(103.35±12.46)nmol/L](P<0.01),而CGRP治疗组ET及IFCa含量与正常对照组比较差异无显著性(P>0.05)。【结论】CGRP能拮抗ET收缩血管的作用,改善脑循环,抑制ET的合成及释放,同时能阻止脑细胞钙内流,保护缺氧缺血性脑损伤。 【Objective】 To study the influence of calcium gene-related peptide (CGRP) on endothelin (ET) and free calcium concentration in brain cells of hypoxic-ischemic neonatal SD (Spraque-Dawley) rats. 【Methods】 The 45 SD rats of 7 days were divided into three groups randomly. In CGRP group, the rats were given CGRP intraperitoneally for 3 days in doses of 3μg/(kg·d) putting into natural salt 2 ml once the hypoxic-ischemic encephalopathy (HIE) model was made; In natural salt group, the rats were given natural salt in doses of 2 ml/d intraperitoneally for 3 days and the rats were treated with pseudo operation in normal group. All the rats were decapitated after three days, and the concentration of ET and free calcium in brain cells were measured with radioimmunoassay and calcium fluorescence indicator in Fura-2 Am. 【Results】 The concentration of ET and free calcium in brain cells of natural salt group [(93.41±12.73)ng/L and(129.47±17.21)nmol/L respectively] were higher than those of normal group [(57.58±6.31)ng/L and(93.72±24.16)nmol/L] and CGRP group [(61.68±6.83)ng/L and(103.35±12.46)nmol/L] significantly (P〈0.01), but there was no significant difference between CGRP group and normal group (P〉0.05). 【Conclusions】 CGRP can antagonize the vasoconstriction of ET and improve brain circulation,so it inhibits synthesis and release of ET. In addition, it can prevent calcium from flowing into brain cells, and protect hypoxic-ischemic cerebral injury.
