摘要
目的观察环境中氧含量对口腔链球菌过氧化氢产生速率的影响。方法采用ABTS-HRP微量板法测定在不同氧含量条件下口腔链球菌过氧化氢产生的速率。结果口腔链球菌在严格厌氧条件下过氧化氢产生速率为9.29nmol/(min×109细胞);当氧含量增高,口腔链球菌过氧化氢合成速率加快;在不同氧含量的环境中口腔链球菌产生过氧化氢的速率差异存在显著性:有氧振荡培养>有氧静置培养>厌氧(P<0.05)。结论环境中氧含量是影响口腔链球菌过氧化氢产生速率的重要因素。
Objective To investigate the influence of environmental conditions with various oxygen concentration on initial rates of hydrogen peroxide produced by S. oralis. Methods After S. oralis were cultivated in anaerobical, aerobically static or shaking condition, respectively 1 ml aliquots of 10^9cells/ml mid-logarithmic phase S. oralis in TSBY were centrifuged and washed by phosphate buffer containing 0. 01~10mmol/L glucose (pH 7.0). After S. oralis were cultured in buffer for 10, 20 and 30minat 37 C, the concentration of hydrogen peroxide in supernatant were assayed speetrophotometrieallyin 96-well micro-plate by ABTS-HRPat A40S. Result Synthesis rate of hydrogen peroxide by S. oralis grown anaerobically was 9. 29 nmol H2O2/(min×10^9cells), there were statistically significant differences among the three groups : aerobically shaking 〉 aerobically static 〉 anaerobiea/ condition(P〈0.05). Conclusion Oxygen was the important influence factor on synthesis rate of hydrogen peroxide by S. oralis.
