摘要 目的研究肿瘤坏死因子相关凋亡诱导配体受体(TRAILR)在肺癌细胞株A549中的表达及TRAIL对肺癌细胞的杀伤作用,探讨TRAIL治疗肺癌的可行性。方法采用RTPCR检测非小细胞肺癌细胞株A549TRAILR的表达。采用不同浓度TRAIL处理A549,MTT法检测药物处理前后肿瘤细胞的凋亡发生率。采用不同浓度TRAIL与不同浓度的化疗药物、中药联合作用于A549,观察其疗效。结果肺癌细胞株A549中有DR5、DR4、DcR2的表达,但DcR1表达缺失。经TRAIL(100μg/L)处理48h,肺癌细胞凋亡发生率约5%。TRAIL联合化疗药物、中药可显著增强A549的凋亡率。结论A549中存在TRAILR类型的表达差异。TRAIL可诱导A549凋亡,联合化疗药物、中药可显著增强A549的凋亡率。 Objective To observe the expression of tumor necrosis factor related apoptosis-indueing ligand receptor (TRAILR) and killing effect of TRAIL on lung cancer cells line A549, in order to investigate the therapeutic potential of TRAIL in lung cancer cells A549. Methods The expression of TRAIL receptors was detected in lung cancer cell line A549 by using PCR. After the A549 cells were treated with different concentrations of TRAIL, the apoptotic rate of the tumor cells was measured before and after treatment by MTT method. After the A549 cells were exposed to different concentrations of TRAIL and chemotherapeutic agents in combination with Chinese medicine, the curative effectiveness was observed. Results DR5,DR4 and DcR2 but not DcR1 were expressed in A549. The apoptotic rate of lung cancer cells, treated with TRAIL (100 μg/L) for 48 h, was about 5 %. TRAIL in combination with chemotherapeutic agents or Chinese medicine could significantly enhance the rate of apoptosis. Conclusion Discrepancy of TRAILR types exited in lung cancer cell A549. TRAIL can induce apoptosis of A549. Combined use of TRAIL and chemotherapeutic agents or Chinese medicine can enhance the rate of apoptosis significantly.
