摘要
目的:利用缺血再灌注大鼠模型研究代谢型谷氨酸受体1(mGluRl)和代谢型谷氨酸受体5(mGluR5)的mRNA表达水平在缺血再灌注后的变化,探讨其在缺血后脑损伤中的作用。方法:采用线栓法制备缺血再灌注大鼠模型,运用RT-PCR技术半定量分析脑组织内mGluRl和mGluR5的mRNA表达水平。结果:以β-Actin为参照,实验组缺血2h再灌注24h后缺血侧mGluRl和mGluR5的mRNA相对值较假手术对照组均显著上升(P<0.05);非缺血侧两者的mRNA相对值与假手术对照组相比,无显著差异(P/0.05)。结论:代谢型谷氨酸受体l和代谢型谷氨酸受体都参与了缺血再灌注后的病理过程。
Objective: To investigate the mRNA changes of metabotropic glutamate receptor 1 (mGluR1) and metabotropic glutamate receptor 5 (mGluR5) in rats with focal cerebral ischemia and reperfusion. Methods: The rat model with focal cerebral isehemia and reperfusinn was induced by an intraluminal filament occlusion and withdrawal of the suture. The gene expressions of mGluR1 and mGluR5 were mearsured by RT-PCR. Results: The expression of mGluR1 and mGluR5 increased in ischemia hemispheres of experiment group compared with hemispheres of sham surgery group (P〈0.05) . There were no difference between non ischemic hemispheres of experiment group and hemispheres of sham surgery group. (P〉0.05) Conclusion: Activation of mGluR1 and mGluR5 has been implicated in the pathophysiology of cerebral ischemia and reperfusion.
