克雷伯杆菌反复感染诱发大鼠肺内c-fos mRNA转录和蛋白表达 被引量:1

摘要 目的:探讨肺炎克雷伯杆菌反复感染在慢性阻塞性肺疾病(COPD)形成中的机制。方法:将大鼠随机分为对照组(C)和肺炎克雷伯杆菌感染组(K)。动态观察肺感染K组肺细胞损伤改变。以免疫组化和原位分子杂交方法分别检测肺内各细胞c-fos蛋白和mRNA表达的变化。结果:K组第8周少部分细支气管管壁有平滑肌细胞和纤维组织增生,第16周纤维组织明显增生穴P<0.01雪。K组感染1周后,在不同时间内肺内各细胞均有c-fos蛋白和mRNA表达增加(P<0.01)。并且各自c-fosmRNA转录水平和c-fos蛋白表达水平基本平行,其中巨噬细胞最早表达c-fos蛋白。从第1天持续到感染第16周。结论:肺炎克雷伯杆菌感染在体内能诱发肺内各细胞损伤和c-fos转录和表达上调,在加重COPD形成中有重要作用。 Objective: To study the lung c-los mRNA transcription and protein expression inuced by Klebsiella Pneumoniae infection in rats, and discuss its pathogenesis in chronic obstructive pulmonary disease (COPD). Methods: Rats were randomly divided into two groups including control group (group C) and Klebsiella Pneumoniae infection group (group K).Lung cell injury changes were observe dynamically in group K. c-los mRNA transcription and protein expression of lung varied cells were measured by immunohistochemistry and hybridization. Results: h few airway bronchiole smooth muscle ceils and fibrous tissue proliferated slightly 8 weeks ater infection, and fibrous tissue obviously proliferated 16 weeks after infection in group K (P 〈0.01). c-fos protein an mRNA expression of different time significantly increased 1 week after infection (P 〈 0.01) and c-fos mRNA transcription and protein expression levels were basically parallel, c-fos protein was first expressed by Phagocyte from 1st day to 16 th weeks after inection (P 〈 0.01). Conclusion: Klebsiella Pneumoniae infection can cause lung varied ceils injury and increase c-fos protein and mRNA expression, and it play an important role in aggravating COPD.
